Someone who is Not Amused is probably annoyed or mad at something stupid, dumb, and/or bad. Chances are that the thing they are Not Amused at was trying to be amusing and failed miserably. PPC agents are often Not Amused at the antics of Mary Sues.
The capital letters here are used to distinguish this particular state of annoyance from simply not finding something amusing.
